Introduction

Western blotting combines the resolution of polyacrylimide gel electrophoresis and the specificity of immunodetection; therefore, providing quick, sensitive, and accurate results. Before employing this procedure it is necessary to have the proper reagents to ensure quality results.

These include a specific monoclonal or polyclonal antibody for the denatured protein of interest, a good secondary antibody conjugated to a stable enzyme (usually alkaline phosphatase or horseradish peroxidase), which yield a colored or luminescent product after the use of a quality substrate for the conjugated enzyme.

Other enzymes have been used as well, but they have not gained widespread acceptance because of limited substrate options. These include s-galactosidase, acetylcholinesterase and catalase. A large selection of substrates is available for performing the Western blotting with an HRP or AP conjugate. The choice of substrate depends upon the necessary sensitivity level of the detection and the instrumentation available for detection (spectrophotometer, fluorometer or luminometer).
