VX-702, one of a series of second-generation, orally active p38 MAP kinase inhibitors, for the potential treatment of inflammation, rheumatoid arthritis and cardiovascular diseases. In June 2005, a phase II clinical trial of VX-702 was initiated in rheumatoid arthritis. VX-702 dose-dependently inhibited the production of IL-6, IL-1? and TNFa (IC50 = 59, 122 and 99 ng/ml, respectively). Pre-incubation of platelets with VX-702 (1 ?M) completely or partially inhibited platelet agonistinduced p38 activation (IC50 = 4 to 20 nM). Soluble in DMSO. Store cool and dry at -20C.
