Biochemically, UbcH1 can catalyze the formation of K48-linked ubiquitin chains in the absence of an E3 ligase. This enzyme has a unique C-terminal UBA domain that plays a critical role in E1 selectivity and chain synthesis. Recently, UbcH1 (or HIP2) has been shown to bind to the N-terminus of Huntington and may play a role in Huntington disease.
