causes accumulation of phosphorylated ERK and inhibits its activity in vivo and in vitro by inhibition of ERK dephosphorylation. In vivo administration SU1498 prevents the pathophysiological symptoms of occupational asthma in Murine Model of Toluene Diisocyanate-Induced Asthma4 and reduces the expression of MMP-9 in Asthma.5

(A) and (B): HUVECs were tested in a Chemokinetic Migration Assay using 8 mm-pore transwell inserts with or without 20 ng/ml VEGF in the lower chamber as chemoattractant, in presence or absence of 20 mM SU 1498 in both chambers. (A) demonstrates the percentage of migrated cells relative to control; average of two different experiments (six microscopic fields each) and (B) shows the photomicrograph of each treatment. (C) and (D) show Western blot analysis of HUVECs treated as described above, probed with anti-phospho-p38 MAPK (C) and with anti-b actin (D). |

Tyrphostin SU 1498 (SU 1498 ) is a potent and selective inhibitor (IC50 = 700 nM) of vascular endothelial growth factor receptor 2 (VEGFR 2) (also known as Fetal Liver Kinase 1 (Flk1) or Kinase-Insert Domain-Containing Receptor ). This receptor tyrosine kinase is expressed exclusively in endothelial cells and has an extensive role in angiogenesis. Inhibition of Flk1 by SU 1498 arrests angiogenesis in in vivo and in vitro models.1 SU 1498 is a very weak inhibitor of PDGFR-kinase, EGFR-kinase and HER-2 kinase.2,3 It has been demonstrated that SU1498
