IMMUNOGEN: Synthetic peptide mapping to the C-terminus of human Tau.

FORMULATION: 100 ug (0.2 mg/ml) purified goat polyclonal antibody in phosphate-buffered saline (PBS) containing 50% glycerol, 1% BSA, and 0.02% thimerosal.

STORAGE CONDITIONS: Store at -20oC. For long term storage, aliquot and freeze at -70oC. Avoid repeated freeze/defrost cycles.

BACKGROUND: Tau, a microtubule-binding protein which serves to stabilize microtubules in growing axons, is found to be hyperphosphorylated in paired helical filaments (PHF), the major fibrous component of neurofibrillary lesions associated with Alzheimer's disease. Hyperphosphorylation of Tau is thought to be the critical event leading to the assembly of PHF. Six Tau protein isoforms have been identified, all of which are phosphorylated by GSK3. This presents the possibility that miscues in GSK3 signaling contribute to the onset of Alzheimer's disease.

SPECIFICITY: The antibody recognizes Tau of human, mouse, and rat origins. Reactivity to other species has not been tested. Mouse brain tissue lysate can be used as a positive control.

APPLICATION: The antibody can be used in Western blotting (1-4 ug/ml), immunoprecipitation (10-20 ug/ml) and immunohistochemistry (15-25 ug/ml). However, the optimal concentrations should be determined individually.
