Introduction:

Human umbilical cord blood, or "cord blood," contains an unusually rich supply of stem cells. These stem cells produce the red, white and immune cells that make up your blood. It plays the role of the blood and immune system and potentially the nervous system, skin and bone tissue as well as the heart, pancreas and other central body tissues. Recently, techniques have been developed for the in vitro culture of stem cells, providing unprecedented opportunities for studying and understanding human embryology. Human stem cells hold out the potential of almost unimaginable cures for a myriad of diseases. That's because they can develop into most of the specialized cells and tissues of the body.

This kit provides a method based on CD45 Side Scatter gating combined with CD34 staining, which is well adapted to the detection of rare events such as CD34+ cells. CD45 is a major component on the lymphocyte membrane surface. The enumeration of CD34+ cells by flow cytometry is commonly employed to assess progenitor/stem cell numbers in peripheral blood, cord blood, and apheresis products used for peripheral blood stem cell transplantation (PBSCT). By flow cytometry is quite simple to identify and enumerate the CD34+ cells, present in physiological conditions on 1-3% of normal bone marrow, 0.1-0.5% of cord blood and 0.001-0.01% of peripheral blood cells. Total nucleated white blood cells (WBCs) are quantified by staining with fluorescein isothiocyanate (FITC)-conjugated CD45 antibody. Simultaneous staining by phycoerythrin (PE)-conjugated CD34 antibody defines an approximate number for the CD34+ progenitor/stem cell sub-fraction.
