In two-electrode voltage clamp of Xenopus oocytes expressing Kv1.2 channels, application of rTityustoxin-Ka inhibits the current amplitude in a concentration dependent (see figures A and B, IC50 of less than 1 nM) and reversible manner. The block spanned the voltage range of these channels activity (figure C). |

| A: Kv1.2 channels currents (in 5 mM K+) elicited by 200 ms depolarization from holding potential of ?00 mV to +20 mV, before and during application of rTityustoxin-Ka of the indicated concentration. B: Mean ?S.D, dose response for rTityustoxin-Ka block of Kv1.2 channels (n was between 3-4 oocytes for each point). C: An example of I-V relations of control and inhibited currents in the same oocyte. |

Tityustoxin-Ka was originally isolated from the scorpion Tityus serrulatus, and was shown to block cloned Kv1.2 with high potency.1 It inhibits stably expressed Kv1.2 channels in fibroblast cell line as assessed by patch clamp with Kd of 0.21 nM.1 Tityustoxin-Ka also reduced the binding of 125I-a-Dendrotoxin to synaptosomal membranes.2
