| The biological activity of rSlotoxin was tested using two-electrode voltage clamp recording from Xenopus oocytes expressing mSlo (a1 subunit alone) channels. 100 nM slotoxin completely abolished the mSlo currents, in a reversible manner. |

| The effect of 100 nM rSlotoxin on heterologously expressed BK currents (mSlo, RNA injected to Xenopus oocytes). Left: Time course of current amplitude changes upon application of the toxin (bars represent the time of toxin perfusion). Right: An example of current response to 100 ms depolarization to +20 mV (from holding potential of ?00 mV) before (red) and during (black) perfusion of 100 nM toxin. |

Native Slotoxin was originally isolated from Centruroides noxius scorpion venom.
Native Slotoxin blocks the Maxi K+ (BKCa or slo, KCNMA1, KCa1.1) channels.1
Slotoxin blocks differentially channels formed by the a1 subunit alone and channels
formed by the a1 combined with auxiliary b subunits. In Xenopus oocytes expressing
hSlo (a1 alone), Kd was calculated to be 1.5 nM and the block is complete and reversible.
With the additional co-expression of either the b1 or b4 subunits, the block become
irreversible or causes the channel to be almost insensitive to the toxin, respectively.1
