Lq2 was originally isolated from Leiurus quinquestriatus hebraeus scorpion venom.1

rLq2 is a recombinant peptide expressed in and extracted from E. coli and purified to homogeneity.

Lq2 blocks Kir1.1 K+ inward rectifier channels (Ki of 410 nM).2

In the planar lipid bilayer, it also blocked single KCa channels from rat muscle with intrinsic KD of 43 nM,1

by introducing brief closures. Lq2 action is on the extracellular side of the pore.

M.W.: 4353

Sequence: QFTQE SCTAS NQCWS ICKRL HNTNR GKCMN KKCRC YS

Purity: >98%

Bioassay:
In two-electrode voltage clamp of Xenopus oocytes expressing Kir1.1 channels, application of rLq2

inhibits the current amplitude in a concentration dependent (see figures A and B, IC50 of 5 nM)

and reversible manner. The block spanned the voltage range of these channels activity (figure C).

A. Kir1.1 channels currents elicited by 200 ms depolarization from holding potential of

-100 mV to +20 mV, before and during application of rLq2 of the indicated concentration.

B.
Mean ?S.D, dose response for rLq2 block of Kir1.1 channels (n was between 4-6 oocytes for each point). The smooth line is a fit giving Ki of 5 nM.

C. Mean ?S.D, I-V relation of control and inhibited currents (n=4).
