Native Heteropodatoxin-2 was originally isolated from the Heteropoda venatoria spider venom.

rHeteropodatoxin-2 is a recombinant peptide expressed in and extracted from

E. coli and purified to homogeneity. Native Heteropodatoxin-2 blocked Ito1 in ventricular myocytes

in a voltage dependent manner with IC50 (at ?FONT size=2>10 mV) of 16 nM.

The toxin also blocked KV4.2 expressed in Xenopus oocytes in a similar manner.1

M.W.: 3414

Sequence: DDCGK LFSGC DTNAD CCEGY VCRLW CKLDW

Purity: >98%

Bioassay:
The biological activity of rHeteropodatoxin-2 was tested using two-electrode voltage clamp recording from Xenopus oocytes expressing the KV4.2 channels.

rHeteropodatoxin-2, inhibition of KV4.2 channels expressed in Xenopus oocytes.

Middle: Current responses to 100 ms depolarization to 0 mV from holding potential

of ?00 mV, delivered every 10 seconds before (red) and during application of 500 nM

rHeteropodatoxin-2.

Left: Time course of amplitude inhibition and wash.

Right: I-V relationships for KV4.2 before and during rHetropodatoxin-2,

perfusion. Also included, the fraction of inhibited current as function of the

voltage step (see inset for symbols) to demonstrate the voltage dependency of block.
