Recombinant Type I Porcine Pancreatic Elastase High-Purity

SPECIFICATIONS.

Chromatographically purified. Lyophilized in .01 M phosphate buffer pH 7.4 containing 0.14 M NaCl,  water soluble powder.  The ultra-high purity recombinant type I porcine pancreatic elastase is homogenous in SDS polyacrylamide gel electrophoresis.

LOW ENDOTOXIN; 0.002 EU/mg.

The endotoxin level in the porcine pancreatic elastase is determined  using a Kinetic-QCL^® Kinetic Chromogenic LAL Assay (Lonza Cat # 50-650U) as instructed by the manufacturer. 

TRYPSIN; residual trypsin activity is 14 ng trypsin per mg elastase.

Store at 5° C or -80° C.  Stable.

67 units per mg using 4.5 mg/ml 10mM of N-Succinyl-ALA-ALA-ALA-p-Nitroanalide (EPC Cat. #NS945) in an elastase activity assay at pH 8.3 at 30^oC. 

Approximately 137-149 units per mg of protein on the substrate elastin-remazol assay described in Methods. Very low trypsin as determined by assay using N-Benzolyl-Phe-Val-Arg-pNA (BENZ) (EPC Cat. #NB32) as the substrate.

PREPARATION.

Recombinant type I porcine pancreatic elastase is expressed in yeast and chromatographically purified on methacrylate at pH 5.0 by elution with salt gradient to obtain the highest specific activity. Recombinant type porcine pancreatic elastase is  produced entirely without exposure to animal tissue or animal-derived products. 

Free of chymotrypsin, lipase, alpha-amylase, carboxypeptidases and other pancreatic enzymes or proteins.
