Charybdotoxin was originally isolated from the venom of the Israeli scorpion
Leiurus quinquestriatus hebraeus. Charybdotoxin blocks KCa1.1 (large conductance Ca2+ activated K+, Slo) channels in nanomolar concentrations2 as well as Kv1.2 (Kd, 14 nM) and Kv1.3 (Kd, 2.6 nM) channels.3 However, experiments with cloned KCa1.1 channels demonstrate the strong effect of the slob subunits on the potency of block by Charybdotoxin (see for example4).

Bioassay: |
This toxin was tested to confirm its ability to potently inhibit Kv1.3 K+ channels expressed in Xenopus oocytes. The IC50 in this assay was 1nM.5 |
 |
The effect of 100 nM rCharybdotoxin on KCa1.1 (left) and Kv1.3 (right, co-expressed with Kvb1.1) channels, expressed in Xenopus oocytes. In both cases the currents were recorded with standard ND96 solution (see above), Membrane holding potential was ?00 mV, stepped every 15 s. to +20 mV for 100 ms. The vertical bar represents 0.1 and 3 mA for KCa1.1 and Kv1.3 respectively. |
