Dose dependent enhancement of TTX-Sensitive NaV currents by rATX-II. (see above) Left: Time course of NaV current amplitude responses to voltage step stimulation in ND7-23 cells. Under patch clamp whole cell, holding potential was set to ?00 mV and 40 ms pulses to ?0 mV were delivered every 10 seconds. Current amplitude was measured 5 ms after the onset of the test pulse (during inactivation). Increasing concentrations of rATX-II (#RTA-700) were perfused and washed in the following order: 50 nM, 250 nM, 500 nM, 1 mM, 2 mM and 5 mM, these periods are indicated by the bars. Middle: Superimposed traces of current responses before (green) and during (black) application of 1 mM rATX-II. Right: A summary dose response curve taken from four experiments such as the one described on the left (mean ?SD). |

Potent enhancement of NaV1.5 channel currents by rATX-II. (see above)
Right: Dose response curve for rATX-II acting on NaV1.5 currents (mean ?SD, n=7-12 experiments for each point) expressed as fold increase in current amplitude, measured 5 ms after the onset of the test pulse (inactivation period). Left: Example traces of NaV1.5 channel currents responses to a 40 ms test pulse (-30 mV, holding potential ?00 mV) before (blue) and during (purple) application of 10 nM rATX-II (#RTA-700). Currents were recorded from HEK-293 cells, stably expressing NaV1.5 channels. |

rATX II is a 47 amino acid peptidyl toxin, originally isolated from Anemonia sulcata sea anemone venom. It is a potent neurotoxin, which modulates voltage-gated Na+ channel gating kinetics by delaying its inactivation and prolonging the action potential of excitable membranes. ATX II has been used as a powerful activator of TTX-sensitive and insensitive Na+ channels in various excitable tissue and cell types (at concentration range of 10-100nM).1-9
