rAgitoxin-2 was tested to confirm its ability to inhibit potently Kv1.3 K+ channels expressed in Xenopus oocytes.
Left: Time course of Kv1.3 current amplitude in response to test pulse of 100 ms to 0 mV (holding potential -100 mV). increasing concentrations of rAgitoxin-2 were perfused at the period marked by the horizontal bars with the concentration marked in nM.
Right: Traces of Kv1.3 currents before and during application of the toxin at increasing concentrations (indicated in the index).

Native Agitoxin-2 was originally isolated from the venom of the Israeli scorpion L. quinquestriatus hebraeus.1 Agitoxin-2 inhibited the native Kv1.3-like current in cultured microglia by 72% at 5 nM and Kv1.3 in MLS-9 cells by 87%.2 rAgitoxin-2 is a potent blocker of the Shaker voltage-gated K+-channels as well as the mammalian homologues of Shaker.1
