Prixotoxin-1 (#P-710) inhibits the K+ current of KV4.2 expressed in Xenopus oocyte. |

| Legend: KV4.2 channel currents were recorded from Xenopus oocytes using the two electrode voltage clamp, with ND 96 solution in the bath. Traces of current response to 100 mSec depolarization from holding potential of ?00 mV to -10 mV, applied every 10 s. in presence or absence of 100 nM phrixotoxin 1. |
Phrixotoxin 1 (PaTx1) is 29 amino acids peptidyl toxin isolated from venom of the theraphosid Phrixotrichus auratus, which potently block A-type K+ currents.
PaTx1 specifically block the Kv4.3 and Kv4.2 currents with an 5 nM < IC50 < 70 nM, by modifying the gating properties of these channels.1
As otherspider toxins, PaTx1 present an active molecular surface, including a hydrophobic patch surrounded by charged residues, which are important for their binding on Kv channels. Electrophysiological studies showed that the PaTx1block Kv4 channels via a mechanism similar to that of hanatoxinson Kv2 channels, as they bind and stabilize the preferentially to the closed stateof the channel, in a voltage-dependent manner.2
PaTx1 have been crucial tool for understanding the contribution of Kv4.3 and Kv4.2 in cardiac electrogenesis. Treatment of mice with PaTx1 resulted in numerous transient cardiac adverse reactions including the occurrence of premature ventricular beats, ventricular tachycardia, different degrees of atrioventricular (AV) block and prolongation of the QT interval.1
Alomone Labs is pleased to offer Phrixotoxin-1, a ntural peptide purified to homogeneity from the venom of the tarantula phrixotrichus auratus.
