p53 Antibody

U937, K562, THP-1 are null (TDH 6-23-10) and Saos-2 are null (TDH 7-15-10).

A NOTE ON p53 HALF-LIFE: There is a large body of literature regarding p53 half-life, which is very dynamic. p53 activity and stability is impacted by oxidative stress, osmotic shock, ribonucleotide depletion, as well as DNA damage. Cellular stressors stimulate a complicated network of signals to induce p53 gene expression (via an autocrine loop) and stability but also can lead to proteosomal degradation of p53. The half-life of p53 can be increased from ~20 minutes to several hours depending on the state of the cells. Please see Maltzman W., Czyzyk L. (1984) Mol. Cell. Biol. 4:1689–1694 as an example.

Product Usage Information

For optimal ChIP results, use 5 μl of antibody and 10 μg of chromatin (approximately 4 x 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

p53 Antibody recognizes endogenous levels of total p53 protein. This antibody binding has been mapped to the amino terminus and DNA binding domain of human p53 protein. This antibody does not cross-react with with other p53-related proteins.

Polyclonal antibodies are produced by immunizing animals with a full-length human p53 fusion protein. Antibodies are purified by protein A and peptide affinity chromatography.
