The biological activity was tested to confirm the ability of K252b to inhibit PKC. |
 |
K252b inhibits the serine phosphorylation of Myristoylated alanin-rich protein kinase C substrate (MARCKS). C6 glioma cells were grown to 70% confluency and then serum starved for 18 h. The cells were preincubated for 30 min in different concentrations of K252b for 20 min and stimulated with TPA (2mM) for 30 min. Cell extracts were prepared for Western blotting. The proteins were probed with anti-Phospho (ser)- MARCKS. |
 |
K252b inhibits tyrosine phosphorylation of TrK A induced by NGF. PC12 cells were incubated in the presence or absence of 10 ng/ml NGF with or without various concentrations of K252b. The inhibitory effect of K252b on neurite outgrowth stimulated by NGF is presented in the picture. |

K252b is an alkaloid isolated from Nocardiopsis sp. soil fungi. It is a less potent derivative of K252a,3-5 cell-permeable protein kinase inhibitor.
This compound potentiates neurotrophin-3 activity in certain neurons by an unknown mechanism. In other neurons it serves as a control to K252a since it accumulates into the plasma membrane of the neuron.1
