Introduction
Free radicals have been implicated in over a hundred disease conditions in humans, including arthritis, hemorrhagic shock, arteriosclerosis, aging, ischemia and reperfusion injury of many organs, Alzheimer and Parkinson's disease, gastrointestinal dysfunctions, tumor promotion and carcinogenesis, and AIDS. Flow cytometry use 2',7'-dichlorofluorescin diacetate (DCFH-DA) 〠dihydroethidium and nitric oxide sensor dye to detect free radical production. The intracellular fluorescent probe 2',7'-dichlorofluorescin diacetate (DCFH-DA, sensitive to H 2 O 2 and hydroxyl radicals) was used to assess intracellular ROS. The oxidation of 2',7'-dichlorofluorescin (DCFH) to the fluorescent 2',7'-dichlorofluorescein (DCF) was investigated by fluorescence, absorption, and electron spin resonance spectroscopy (ESR). Set up the cytometry for the standard measurement of linear FL1 fluorescence, using a 530 ± 30 nm band –pass filter for detection of DCF fluorescence.
