Western Blot: Approx 30kDa band observed in Mouse Heart lysates (calculated MW of 28.8kDa according to NP_071878.2). An additional band of unknown identity was also consistently observed at 15kDa. This band was successfully blocked by incubation with the immunizing peptide. Recommended concentration: 1-3µg/ml. Primary incubation 1 hour at room temperature. Preliminary testing was unsuccessful on Rat Kidney for this particular batch.Immunofluorescence: Strong expression of the protein seen in the cytoplasm of NIH3T3 cells. Recommended concentration: 10µg/ml.

Flow Cytometry: Flow cytometric analysis of NIH3T3 cells. Recommended concentration: 10ug/ml.

Peptide ELISA: antibody detection limit dilution 1:64000.

Description (Long Description) : 

NAD-dependent protein deacetylas. Activates or deactivates mitochondrial target proteins by deacetylating key lysine residues. Known targets include ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA and the ATP synthase subunit ATP5PO. Contributes to the regulation of the cellular energy metabolism (PubMed:24252090). Important for regulating tissue-specific ATP levels. In response to metabolic stress, deacetylates transcription factor FOXO3 and recruits FOXO3 and mitochondrial RNA polymerase POLRMT to mtDNA to promote mtDNA transcription. Acts as a regulator of ceramide metabolism by mediating deacetylation of ceramide synthases CERS1, CERS2 and CERS6, thereby increasing their activity and promoting mitochondrial ceramide accumulation
