| Antibody Usage Summary: | Immunohistochemistry: LS-B2808 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody LS-D1, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2808 was determined to be 5 ug/ml. Western Blot: Multiple splice variants have been reported for this protein. Detection of Gli-2 by western blot may be enhanced if nuclear extracts are used instead of whole cell lysates as the expression/abundance of Gli-2 is likely to be low. Furthermore, Gli-2 expression is likely to be developmentally regulated and induced, making it difficult to detect in whole tissue homogenates. |
