Compounds and membranes are preincubated at 4deg;C for 15 min prior to initiating the binding reaction by the addition of radioligand. The assay run under these conditions showed a 90 to 96% specific binding signal. The reaction is incubated on an orbital shaking platform for 1 h at room temperature at a speed of 3 revolutions per min. The MultiScreen-FB opaque 96-well filtration plates containing glass fiber filters are used to terminate the binding reactions and to separate bound from free radioligand. The plates are presoaked with 0.5% polyethylenimine in water for a minimum of 2 h at room temperature to reduce nonspecific binding of [span style=margin: 0px; padding: 0px; position: relative; vertical-align: baseline; top: -0.5em;3/spanH]nisoxetine during the harvest of the binding reactions. The PEI solution is aspirated using a vacuum manifold from the filter plates just before completion of the reaction incubation time. Aliquots of each reaction (180 mu;l of each 200-mu;l reaction well) are transferred from the reaction plates to the filter plates using a Zymark Rapid Plate-96 automated pipette station. The binding reaction is terminated by vacuum filtration through the glass fiber filters. The filter plates are aspirated at 5 to 10 inches of Hg, and the wells are washed seven times with 200 mu;l of wash buffer (50 mM Tris-HCl, 0.9% NaCl, pH 7.4, 4deg;C) using a 12-channel aspiration/wash system. Plastic bottom supports are removed from the filter plates, and the plates are placed in plastic liners. A 50-mu;l aliquot of scintillation fluid is added to each well, and the top of each plate is sealed with adhesive film. The plates are vigorously shaken on an orbital shake table at 5 rpm for 10 to 15 min to ensure adequate equilibration of aqueous-to-solvent partitioning. The collection of raw counts per minute data are done using a Wallac Microbeta counter./td
