Calyculin A was initially isolated from the marine sponge Discodermia calyx.¹ It is a very potent, 

cell-permeable PP-1 and PP-2 inhibitor (IC₅₀: 2nM and 0.5 -1nM respectively),² and extremely specific  

for PP-1 and PP2A among other acidic, alkaline and protein phosphatases.^2,3

Bioassay:

The activity was tested to confirm its ability to inhibit cell proliferation in MCF7 cells.

Calyculin A inhibits the growth of breast cancer epithelial MCF7 cells. MCF7 cells were plated at 8 x 103 cells/well in 96 well plate with different concentrations of Calyculin A for 4 days. The cell quantity was measured using methylene blue method. The number of cells were normalized with respect to control (100%) and plotted against Calyculin A concentration.
