At the day of the binding experiment, the membranes are defrosted and homogenized. The protein concentration is determined using the Bradford protein assay and the membranes are diluted in the assay buffer down to 65 mu;g protein/mL (50 mM Trisndash;HCl, 10 mM MgClspan style=margin: 0px; padding: 0px; position: relative; vertical-align: baseline; top: 0.25em;2/span, 1 mM EGTA and 10 mu;M pargyline, pH 7.4).
