Use & Storage
Depending on desired assay signal and conditions, biotin-Ub should be used in conjunction with native ubiquitin with a 1:1 to 1:5 ratio of biotin-Ub: native Ub, respectively, with a total combined concentration range of 100-500 μM. Typical amounts for a 20 μL reaction: 5 μg - 15 μg biotin-Ub, 20 μg native ubiquitin. Assessment of ubiquitination performed via avidin-linked detection.
Store at -20°C once reconstituted. Avoid multiple freeze/ thaw cycles.

Ubiquitin modified with biotin via primary amine coupling. This results in multiple biotinylated ubiquitin species modified at the N-terminus, as well as lysine residues. Although having a fully functional C-terminus, lysine modification may limit the ability of this reagent to propagate polyubiquitin chains. Biotinylated ubiquitin can be detected using avidin-linked reagents.
