Description

Anti-CaVb3

Product#: ACC-008

Sizes:
50 µl
0.2 ml

Host:
Rabbit

Epitope:

Peptide (C)DRNW QRNRP WPKDS Y, corresponding to residues 463-477 of rabbit CaVb3 (Accession P54286).

Putative epitope location:
Intracellular, C-terminus.

Homology:

Identical in all splice forms.

Applications:

Western Blotting:

Western blotting of rat brain membranes:

1.       Anti-Cavb3 antibody (#ACC-008) (1:200).

2.       Anti-Cavb3 antibody, preincubated with the control peptide antigen.

Immunohistochemistry:
Immunohistochemical staining of rat hippocampus

Immunohistochemical staining of Cavb3 channel with Anti-Cavb3 antibody (#ACC-008) in rat hippocampus.

A, Cavb3 channel (red) appears in neurons (arrows).

B, staining of nerve cells with mouse anti parvalbumin (a calcium binding protein, green) demonstrates the restriction of Cavb3 to cell bodies.

C, Confocal merge of Cavb3 and parvalbumin demonstrates some co-localization of these proteins.

The b3 protein is an auxiliary subunit of the CaV (voltage dependent calcium) channel multimer complex. Four b isoforms were identified in mammalian genoms and they share some sequence as well as structure homology.1-3  The structure of CaVb3 was recentely determined (as well as other CaVb subunits; for review see reference 3). CaVb subunits are cytoplasmic proteins, that bind the cytoplasmic face of the pore forming CaV a1 subunits. These interactions have profound effects on channel trafficing to the plasma membrane as well as on current parameters such as voltage dependent kinetics and interactions with modulatory proteins.1,2  CaVb3 might interact with all the poreforming subunits that have been tested and in vivo is expressed in smooth muscle, trachea, aorta, lung and brain.1

Alomone Labs offers a polyclonal antibody directed against an epitope at the C-terminus of the auxiliary subunit CaVb3.
