Description
ALLERGEN DETECTION KIT
(Cat. No. FC015)

Introduction:
Immediate hypersensitivity is characterized by the allergen-induced mast cell and peripheral blood polynuclear basophiles activation and immediate release of highly active biological compounds stored in vesicles (i.e., histamine) or rapidly produced (i.e., leukotrienes). Flow Cytometry has a reliable and safe in vitro analysis of the capacity of basophiles to release these compounds under specific challenges is very helpful in con-firming the diagnostic and treatment evaluations. the peripheral blood polynuclear basophiles are suitable for in vitro analysis.
Flow cytometry (FCM) have been developed for basophiles identification and activation analysis, though they are rare (0.33–1.35% of leukocytes). We choose IgE and CD63 as basophiles identify marker. Membrane IgE is the most restricted marker for basophiles. CD63, which is normally expressed on the inside of the -vesicle membrane, is a sensitive and reliable membrane marker of basophiles activation. Under membrane IgE cross-linking, the fusion of the vesicle to the plasma membrane allows the expression of CD63 on the external membrane that can be detected directly by FCM.
Compared with the routinely performed tests, Flow cytometry test is a sensitive, automated technique, allows quantitative analysis of single-cell and we could identify the type of cell that was activated with exposure to the allergen.
