The biological activity was tested to confirm the ability of Okadaic Acid but not 1-Nor-Okadaone to inhibit phophatase activity of PP-2A. |

3T3-L1 cells were grown to confluency then serum starved for 18 h. For in vivo phosphatase activity measurement, the growth medium was replaced to HENKî–˜ and the cells were preincubated for 30 min in presence or absence of 100 nM Okadaic Acid (Cat#O-800, Cat#O-900) and 100 nM I-Nor-Okadaone and for 20 min with or without of 20mM NEM. The cells were stimulated with 5 ng/ml PDGF for 30 min and cells extracts were analyzed by Western blotting for Akt using Ser-473 phospho-specific antibodies. |

1-Nor-Okadaone is structural analog of Okadaic acid. This compound is a less active homolog than the Okadaic Acid and Okadaol as inhibitors of Protein Phosphatase 1 and Protein Phosphatase 2A, and is thus widely used as negative control for Okadaic Acid.
